CUT&Tag介绍


CUT&Tag(Cleavage Under Targets & Tagmentation)能够检测特定转录因子的结合位点或特定组蛋白修饰的发生位点。自从2019年Steven Henikoff 在Nature Communications发布该技术以来(目前引用超过1000次),该技术以其相对简化的实验操作,相对宽松的抗体级别要求,逐渐代替了ChIP-seq(Chromatin Immunoprecipitation sequencing)技术,成为表观遗传学研究的核心技术之一。


CUT&Tag通过引导特定抗体和Tn5转座酶复合物到指定转录因子的结合位点/指定组蛋白修饰的发生位点上,从而实现对结合位点DNA的精准切割和标记。这些标记的DNA片段进一步通过PCR扩增和高通量测序,从而提供转录因子/组蛋白修饰在全基因组范围内的结合位点或发生位点信息。CUT&Tag技术在研究基因调控、转录因子结合、组蛋白修饰和染色质结构等方面具有广泛应用。
